Regeneron Pharmaceuticals has finalized a comprehensive research and development collaboration valued at $2.32 billion with Parabilis Medicines. The joint initiative is structured to leverage Parabilis’ proprietary Helicon peptide platform to design innovative therapeutic assets, specifically focusing on antibody-Helicon conjugates (AHCs). From a biochemical perspective, Helicons represent stabilized, cell-penetrating alpha-helical peptides engineered to engage with intracellular protein targets possessing flat surfaces, which typically resist modulation by conventional small molecule formulations.
Under the established financial guidelines, Parabilis is set to receive an initial capital influx of $125 million, consisting of a $50 million upfront milestone fee and a $75 million equity investment from Regeneron during the developer’s upcoming financing round. Parabilis remains eligible to secure up to an additional $2.2 billion in contingent regulatory and developmental milestone payments, supplemented by tiered commercial royalties. The operational framework stipulates that while both enterprises will jointly participate in the discovery phases of new Helicons and AHC architectures, Regeneron will assume sole responsibility for subsequent clinical advancement, manufacturing operations, and international commercialization.
The strategic objective of the combined AHC platform centers on merging the selective targeting precision of proprietary antibodies (derived from Regeneron’s VelocImmune technology) with the therapeutic payload capabilities of Helicon units. This dual-action delivery mechanism aims to precisely modulate specific intracellular proteins of interest, potentially establishing a distinct therapeutic modality capable of addressing historically undruggable targets across multiple medical specialties.
